Protocells are Not Toxic to Raw 264.7 Macrophage Cells

24 well plates seeded with 1 × 10 5 cells were treated for 24 hours with protocells lacking a lipid bilayer (A, B) or a lipid bilayer composed of DOTAP: CHOL 1:1 (C, D) DOPC (E, F) or DOTAP (G, H). Cells were then subjected to a Life/Dead cell viability assay (Invitrogen, Inc.). Live cells, stained green (Calcein-AM) (A, C, E, G), made up 95% of the total population, while dead cells, stained red (Ethidium homodimer) (B, D, F, H), composed only 5%. Confocal imaging at 10X.
